Overview (Skills)

•	Manipulation of DNA, RNA, and protein molecules in both prokaryotic and eukaryotic cells.
•	Culturing mammalian cell lines (human primary and cancer cell lines, mouse epithelial, ES and tumor cell lines). 
•	Isolation and growth of immune cells from human PBMCs and mouse tissues/ tumors. 
•	FACS-based phenotyping of immune cells (monocytes, dendritic cells and T cells). 
•	Priming and expansion of tumor and viral antigen-specific T cells using peptide or IVT mRNA-(LNP) as modality.
•	Epitope mapping of dendritic cells and TCRs.
•	Small scale production of in vitro transcribed mRNAs with GMP quality.
•	Production of research scale of high quality vectors (Lenti, Retro, and varieties of non-viral vectors) for the expression of RNAs and Proteins.

•	Transfection of immune cells with plasmid/lenti virus/mRNA.
•	Using CRISPR-based method to generate knock-out or haploid human primary cell lines, mouse ES cells and cancer lines. 
•	Regulation of gene expression using siRNAs, shRNAs, or CRISPR.
•	Analyzing and purification of small RNAs (small nucleolar RNAs), mRNAs (human tumor antigens), and large RNAs (ribosomal RNAs and XIST RNA).
•	Generation of recombinant proteins (GFP/GST-fused or HIS6 and other epitope-tagged). 
•	Creation of hundred human stable cell lines for the expression of proteins and RNAs of interest.
•	Purification of protein-protein and protein-RNA complexes from E. coli, yeast and mammalian cells.
•	Western blot analysis (WB), immunoprecipitation (IP), immunofluorescent staining (IF) of fixed cells, RNA fluorescent in situ hybridization (RNA FISH).
•	Yeast (Saccharomyces cerevisiae) genetics, molecular biology and biochemistry.  
•	Cell culturing and classic genetic manipulation on E. coli, Salmonella typhimurium, Pseudonomus sp.
